Abstract

Abstract A method for overexpressing a bioelastic polypeptide in a host cell is provided. A nucleic acid encoding a bioelastic polypeptide having pentapeptide, tetrapeptide, hexapeptide or nonapeptide repeating units is introduced into a host cell. The host cell is grown under conditions that provide for expression of the polypeptide as at least 40% of the total cellular protein of the host cell.

Claims Analysis

Total Claims: 7
Claim 1
Independent Claim 130 words
A method for overexpressing a bioelastic polypeptide in a prokaryotic host cell, said method comprising: (a) introducing into said host cell, a vector comprising a promoter operably linked to a nucleic acid encoding a bioelastic polypeptide comprising pentapeptide, tetrapeptide, hexapeptide or nonapeptide repeating units; and (b) growing said host cell to provide for expression of said polypeptide in the inclusion bodies of said host cells, where the volume of said inclusion bodies comprises 40-90% (v/v) of the total cellular volume of said host cell and the amount of inclusion bodies correlates to the amount of polypeptide expressed; with the proviso that when said promoter is an inducible promoter said host cells are grown and said polypeptide is expressed in the absence of an inducer normally associated with said inducible promoter.
Claim 2
Claim 3
Dependent Claim 13 words
The method of claim 1 where said prokaryotic host cell is E. coli.
Claim 4
Dependent Claim 26 words
The method of claim 1 wherein the volume of said inclusion bodies comprises at least 60% (v/v) of the total cellular volume of said host cell.
Claim 5
Dependent Claim 26 words
The method of claim 1 wherein the volume of said inclusion bodies comprises at least 80% (v/v) of the total cellular volume of said host cell.
Claim 6
Dependent Claim 24 words
The method of claim 1 wherein the volume of said inclusion bodies comprises 90% (v/v) of the total cellular volume of said host cell.
Claim 7
Dependent Claim 12 words
The method of claim 1 wherein said promoter is an inducible promoter.
