Abstract
Abstract Virus sub-unit vaccines which are substantially free from pyrogens, virion nucleic acids and unwanted antigenic material are prepared by introducing inactivated whole virus into a continuous loading zonal ultracentrifuge provided with a density gradient containing a haemolytic surfactant and binding the split sub-units isopycnically. All ether-sensitive viruses can be used, especially influenza virus.

Total Claims: 17
Claim 1
Independent Claim
89 words
A process for the preparation of a virus sub-unit vaccine derived from an ether-sensitive virus and being substantially free from pyrogens virion nucleic acids and unwanted antigenic material, including the step of introducing a liquid medium containing the inactivated whole virus into a continuous loading zonal ultracentrifuge provided with a density gradient solution containing a haemolytic surfactant, whereby the virions enter the density gradient and are split by the surfactant and the antigenic sub-units are banded isopycnically, the fraction or fractions containing the protective antigen sub-units subsequently being recovered.
Claim 2
Dependent Claim
Claim 3
Dependent Claim
20 words
A process as claimed in claim 1 in which a continuous loading zonal ultracentrifuge of the KII type is used.
Claim 4
Dependent Claim
21 words
A process as claimed in claim 1 in which the density gradient solution is a colution of sucrose in buffered saline.
Claim 5
Dependent Claim
22 words
A process as claimed in claim 1 in which the density range of the density gradient is from 1.02 to 1.24 g/ml.
Claim 6
Dependent Claim
19 words
A process as claimed in claim 1 in which the surfactant is an aryl ether adduct of ethylene oxide.
Claim 7
Dependent Claim
16 words
A process as claimed in claim 5 in which the surfactant is nonyl phenol 9-10 oxyethylene.
Claim 8
Dependent Claim
23 words
A process as claimed in claim 6 in which the overall concentration of the surfactant in the gradient is at least 0.5% v/v.
Claim 9
Dependent Claim
17 words
A process as claimed in claim 7 in which the concentration of the surfactant is about 1%.
Claim 10
Dependent Claim
