Non-radioactive, single-stranded labelled probe, method for its production and method of detecting a target nucleotide sequence using this probe
Abstract
The invention relates to a new probe for detecting a determined nucleic sequence. It consists of a recombinant DNA containing an insert complementary to the nucleic sequence to be detected and nucleic sequences derived from the (+) DNA of phage M13, the bases used in the composition of this recombinant DNA, in particular the thymine groups being replaced by 5-bromo-uracil groups. The invention also relates to a process for the manufacture of this probe, this process comprising the cultivation of cellular hosts, more particularly E. coli, auxotrophs for thymine, in a medium containing a limiting content of thymine and of 5-bromo- uracil. After the required incubation time, the supernatant is removed and the probe is recovered from the bacterial cells.
