Claim 1
Independent Claim
68 words
A method of producing a long-term continuously proliferating mammalian neuroblast cell culture comprising: culturing neuroblast cells in a vessel in a serum-free basal media supplemented with about 10 ng/ml to 100 ng/ml of basic fibroblast growth factor, wherein a surface in the vessel allows attachment of the cells; and wherein the cells proliferate for greater than seven days, thereby resulting in a long-term continuously proliferating neuroblast cell culture.
Claim 2
Independent Claim
68 words
A method of producing a long-term continuously proliferating mammalian neuroblast cell culture comprising: culturing neuroblast cells in a vessel in a serum-free basal media supplemented with about 10 ng/ml to 100 ng/ml of basic fibroblast growth factor, wherein a surface in the vessel allows attachment of the cells; and wherein the cells proliferate for greater than seven days, thereby resulting in a long-term continuously proliferating neuroblast cell culture.
Claim 3
Dependent Claim
33 words
The method of claim 1, wherein the neuroblast cell are derived from neural tissue selected from the group consisting of hippocampus, cerebellum, spinal cord, cortex, striatum, basal forebrain, ventral mesencephalon, and locus ceruleus.
Claim 4
Dependent Claim
25 words
The method of claim 1, wherein the surface in the vessel is treated with a polybasic amino acid to allow attachment of the neuronal cell.
Claim 5
Dependent Claim
12 words
The method of claim 3, wherein the polybasic amino acid is polyornithine.
Claim 6
Dependent Claim
25 words
The method of claim 1, wherein the surface in the vessel is treated with an extracellular matrix molecule to allow attachment of the neuronal cell.
Claim 7
Dependent Claim
21 words
The method of claim 5, wherein the extracellular matrix molecule is selected from the group consisting of laminin, collagen and fibronectin.
Claim 8
Dependent Claim
20 words
The method of claim 1, wherein the neuroblast cell are cultured in serum-containing media prior to culture in serum-free media.
