Abstract

Abstract Method for treating nucleic acid that is to be amplified such that the nucleic acid is, after amplification, subsequently unamplifiable. Nucleic acid amplification is performed in a reaction containing means in the presence of an isopsoralen. The reaction is thereafter irradiated to render the amplified nucleic acid substantially unamplifiable.

Claims Analysis

Total Claims: 4
Claim 1
Independent Claim 167 words
A method for rendering amplified nucleic acid substantially unamplifiable comprising: a) providing in any order reaction components comprising: i) nucleic acid, ii) amplification reagents, iii) at least one polymerase that is capable of being inhibited by isopsoralen monoadducts, iv) one or more isopsoralen compounds, and v) means for containing a reaction; b) adding to said reaction containing means, in any order, said nucleic acid and said amplification reagents, to make a reaction mixture; and c) adding to said reaction mixture without specifying temporal order, i) said polymerase under conditions such that amplification of said nucleic acid can begin, and ii) said one or more isopsoralen compounds; d) closing said reaction containing means after said adding of said polymerase and said isopsoralen compounds to said reaction mixture; and e) exposing said reaction containing means to ultraviolet radiation under conditions such that said isopsoralen compounds are activated so as to form covalent isopsoralen monoadducts with the amplified nucleic acid and said amplified nucleic acid is thereby rendered substantially unamplifiable.
Claim 2
Claim 3
Dependent Claim 16 words
The method of claim 1 further comprising the step: detecting said amplified, substantially unamplifiable nucleic acid.
Claim 4
Dependent Claim 19 words
The method of claim 2 wherein said detecting comprises hybridization between said unamplifiable nucleic acid and a hybridization probe.
