Measuring method for human chorionic gonadotropin
Abstract
Abstract PURPOSE:To realize a measuring method having a little cross-reactivity to progestrone and high specificity, by using a solid phase fixed monochronal antibody for human chorionic gonadotropin. CONSTITUTION:In order to perform the measurement of human chorionic gonadotropin (HCG) by a sandwich method, inspecting material to be measured is added to a solid phase carried out the fixation of a monochronal antibody and is reacted at 4-40 deg.C optional temeprature for several hours and hereafter, reacted solution is washed and is removed. HCG in the inspection material is combined with the antibody fixed on the surface of the solid phase by this reaction. Next, an enzyme labelled antibody is added and combined product composed of the antibody (solid phase side)-HCG-the enzyme labelled body is produced on the surface of the solid phase by reacting at 4-40 deg.C for several hours. Then, an enzyme substrate is added after removing excess reaction solution and an enzyme reaction is carried out and then, the activity of enzyme is measured. Further, the value of HCG in the inspection material is found by comparing with a calibration curve made by using a standard solution preliminarily.
