Abstract

Abstract The invention provides a method for producing a biological material comprising reconstituted cartilage tissue, wherein the method comprises isolating chondrocytes from cartilage tissue, contacting a substrate comprised of a continuous, flexible sheet of demineralized natural bone, having a thickness less than about 1.5m, with said chondrocytes, and culturing the chondrocytes on the substrate in growth media to produce a biological material characterized by a continuous layer of cartilage tissue. The invention also provides a biological material made by this method.

Claims Analysis

Total Claims: 12
Claim 1
Independent Claim 35 words
A biological material comprising cartilage tissue reconstituted on a substrate in vitro from isolated chondrocytes wherein the substrate is a continuous, flexible sheet of demineralized natural bone, having a thickness less than about 1.5 mm.
Claim 2
Independent Claim 67 words
A method for producing a biological material comprising reconstituted cartilage tissue, wherein the method comprises isolating chondrocytes from cartilage tissue, contacting a substrate comprised of a continuous, flexible sheet of demineralized natural bone, having a thickness less than about 1.5 mm, with said chondrocytes, and culturing the chondrocytes on the substrate in growth media to produce a biological material characterized by a continuous layer of cartilage tissue.
Claim 3
Dependent Claim 15 words
The method of claim 2 wherein the chondrocytes are isolated from human articular cartilage tissue.
Claim 4
Dependent Claim 15 words
The method of claim 3, wherein the chondrocytes are isolated by sequential enzyme digestion techniques.
Claim 5
Independent Claim 58 words
A method for making a cartilaginous structure comprising providing a substrate comprised of a continuous, flexible sheet of demineralized natural bone, having a 5 thickness less than about 1.5 mm, in a nutrient environment and attaching cartilage cells to the substrate to form a cartilaginous structure suitable for implantation into a patient to replace defective or missing cartilage.
Claim 6
Dependent Claim 31 words
The method of claim 5 further comprising proliferating the cells on the substrate in vitro in a nutrient media, thereby to produce a structure comprising the expanded cells on the substrate.
Claim 7
Dependent Claim 16 words
A biological material as obtainable by the method of any one of claims 2 to 4.
Claim 8
Dependent Claim 14 words
A cartilaginous structure as obtainable by the method of claim 5 or claim 6.
Claim 9
Dependent Claim 24 words
A biological material of claim 1 or claim 7, or a cartilaginous structure of claim 8 for implantation in a human or animal body.
Claim 10
Dependent Claim 33 words
Use of a biological material of claim 1 or claim 7, or of a cartilaginous structure of claim 8 in the manufacture of an implant for implantation in a human or animal body.
Claim 11
Dependent Claim 25 words
Use according to claim 10 wherein the implant is for implantation without prior in vitro proliferation, on the substrate, of cells attached to the substrate.
Claim 12
Dependent Claim 30 words
Use according to claim 10 wherein the implant is for the repair of cartilage damaged by inflammation, trauma or ageing, or for the repair of cartilage which is congenitally defective.
