Abstract

Abstract Biosensors for qualitative and quantitative analysis comprise an amphipathic liquid crystalline membrane composed of a lipid bilayer attached to a recording electrode via bridging anchoring molecules. The lipid bilayer is doped with biologic or synthetic ion channels and is in continuous contact with a bulk aqueous medium on both its surfaces. The bridging anchoring molecules may contain a phospholipid moiety linked to a polyoxyalkylene chain terminated with a thiol or thioether residue.

Claims Analysis

Total Claims: 10
Claim 1
Independent Claim 86 words
A biosensor for qualitative and quantitative analysis comprising (1) a reference electrode at its upper part, (2) a recording electrode at its bottom, and (3) an amphipathic liquid crystalline membrane composed of a lipid bilayer doped with synthetic or biologic ion channels, said lipid bilayer being attached to the recording electrode via bridging anchoring molecules and being in continuous contact with a bulk aqueous electrolyte medium on both its surfaces and having its boundaries sealed by an apolar contact with the apolar material of the walls.
Claim 2
Dependent Claim 32 words
A biosensor according to claim 1 wherein a solvated phospholipid bilayer is attached to the recording electrode surface via bridging anchoring molecules comprising a hydrophilic spacer arm joined to a phospholipid moiety.
Claim 3
Dependent Claim 38 words
A biosensor according to claim 1 or 2 wherein the bridging anchoring molecules comprise a phosphatidylethanolamine residue linked to a polyoxyalkylene chain terminated by a thiol or thioether residue for attachment to the surface of a metal electrode.
Claim 4
Dependent Claim 42 words
A biosensor according to any of claims 1 to 3 wherein the bridging anchoring molecule has the formula PE-NH-(CH₂-CH₂-O) n -CH₂-CH₂-SH wherein PE-NH is the residue of phosphatidylethanolamine and n is an integer of from about 7 to about 24, preferably 11.
Claim 5
Dependent Claim 40 words
A biosensor according to any of claims 1 to 4 wherein the ion channels are proteins selected from natural receptors and hybrid receptors having the receptive part which interacts with a ligand and the channel-forming part of a second protein.
Claim 6
Dependent Claim 45 words
A biosensor according to any of claims 1 to 4 wherein the ion channels are synthetic mellitin-like peptides having a hapten as the receptive part and an antibody directed against the hapten as an inhibitory ligand component, the hapten being an analyte or analyte-like moiety.
Claim 7
Dependent Claim 18 words
A biosensor according to claim 6 wherein the ion channel is the peptide CH-1 of the formula NH₂-Gly-Trp-Gly-Ala-Val-Leu-Lys-Val-Leu-Thr-Thr-Gly-Leu-Pro-Ala-Leu-Ile-Ser-Cys-Ile-Lys-Gln-amide.
Claim 8
Dependent Claim 80 words
A method for the analysis of a ligand wherein a sample containing the ligand is contacted with a biosensor according to claim 5 wherein the ion channels comprise the receptor to said ligand or comprise at least its receptive part attached to a channel-forming part of a hybrid protein, and the binding of the ligand triggers the opening of the ion channel in the lipid bilayer leading to a change in electric conductivity that is measured by the recording electrode.
Claim 9
Dependent Claim 82 words
A method for the analysis of an analyte wherein a sample containing said analyte is contacted with a biosensor according to claim 6 wherein the ion channels are mellitin-like peptides containing said analyte or analyte-like moiety as the hapten and which are bound to an antibody directed against the hapten, whereby the antibody molecules are released thus triggering the opening of the ion channels in the lipid bilayer leading to a change in electric conductivity that is measured by the recording electrode.
Claim 10
Dependent Claim 15 words
A method according to claim 9 wherein the biosensor is as defined in claim 7.
