Abstract

Abstract Highly purified antihemophilic factor is produced by a process comprising a PEG precipitation step, a gel filtration step and a heat treating virus inactivation step. Al(OH)₃ adsorption and PEG precipitation carried out at room temperature allow processing to proceed directly to a gel filtration step.

Claims Analysis

Total Claims: 10
Claim 1
Independent Claim 78 words
A process for the production of a concentrate of antihemophilic factor (AHF) from cryoprecipitate comprising the steps of: (a) dissolving said cryoprecipitate; (b) removing non-AHF proteins by precipitation with polyethylene glycol (PEG); (c) heat treating said AHF for viral inactivation; then (d) passing said AHF through a gel filtration column containing a size exclusion resin to remove said chemical and to concentrate said AHF to at least 35 units of Factor VIII activity per ml of pooled concentrate.
Claim 2
Dependent Claim 78 words
A process for the production of a concentrate of antihemophilic factor (AHF) from cryoprecipitate comprising the steps of: (a) dissolving said cryoprecipitate; (b) removing non-AHF proteins by precipitation with polyethylene glycol (PEG); (c) heat treating said AHF for viral inactivation; then (d) passing said AHF through a gel filtration column containing a size exclusion resin to remove said chemical and to concentrate said AHF to at least 35 units of Factor VIII activity per ml of pooled concentrate.
Claim 3
Dependent Claim 17 words
The process of claim 1 wherein said resin has a fractionation range from 300 to 15,000,000 daltons.
Claim 4
Dependent Claim 24 words
The process of claim 1 further comprising the step of precipitating AHF with a mixture of glycine and NaCl prior to said gel filtration.
Claim 5
Dependent Claim 16 words
The process of claim 1 wherein PEG is added in an amount of 2 - 5%.
Claim 6
Dependent Claim 27 words
The process of claim 1 further comprising the step of ultrafiltering said concentrate to obtain at least 100 units of Factor VIII activity per ml of concentrate.
Claim 7
Dependent Claim 55 words
A process for the production of antihemophilic factor concentrate (AHF) comprising the steps of: (a) preparing a cryoprecipitate; (b) removing non-AHF proteins by precipitation to form a solubilized AHF pool; (c) passing said pool through a gel filtration column; and (d) heating AHF obtained from said column in a solution in the presence of sucrose.
Claim 8
Dependent Claim 55 words
A process for the production of antihemophilic factor concentrate (AHF) comprising the steps of: (a) preparing a cryoprecipitate; (b) removing non-AHF proteins by precipitation to form a solubilized AHF pool; (c) passing said pool through a gel filtration column; and (d) heating AHF obtained from said column in a solution in the presence of sucrose.
Claim 9
Dependent Claim 15 words
The process of claim 6 further comprising the step of concentrating said pool by ultrafiltration.
Claim 10
Dependent Claim 21 words
The process of claim 7 wherein said heating in the presence of sucrose takes place in 1.2 to 0.6 mg/ml sucrose.
