Abstract

Abstract A semi-automated biological sample analyzer and subsystems are provided to simultaneously perform a plurality of enzyme immuno assays for himan IgE class antibodies specific to a panel of preselected allergens in each of a plurality of biological samples. A carousel is provided to position and hold a plurality of reaction cartridges. Each reaction cartridge includes a plurality of isolated test sites formed in a two dimensional array in a solid phase binding layer contained within a reaction well which is adapted to contain a biological sample to be assayed. The carousel and cartridges contain structures which cooperate to precisely position the cartridges in each of three separate dimensions so that each cartridge is positioned uniformly. An optical reader operating on a principle of diffuse reflectance is provided to read the results of the assays from each test site of each cartridge. Also provided is a subsystem which provides predetermined lot-specific assay calibration data which is useful for normalizing the results of various assays with respect to predetermined common standard values.

Claims Analysis

Total Claims: 8
Claim 1
Independent Claim 128 words
A reaction cartridge for use in a biological sample analyzer including cartridge conveying means having at least one opening for receiving said cartridge, said opening including first locking means; and optical reader means operable to read a plurality of biological test sites in close proximity to each other, said cartridge comprising: support means having a plurality of isolated biological test sites formed therein; reaction well means containing said support means therein and adapted to hold fluids in contact with said test sites, said well arranged to provide direct optical access to said plurality of test sites for reading by said optical reader means; and second locking means for cooperating with said first locking means to position and lock said cartridge in a predetermined three-dimensional position in said opening.
Claim 2
Dependent Claim 128 words
Claim 3
Dependent Claim 33 words
The cartridge of claim 1 wherein said cartridge further includes removable cover means for covering said well means, said cover means having a port for the introduction of fluids into said well means.
Claim 4
Dependent Claim 64 words
The cartridge of claim 2 wherein said cover means comprises a first cover layer bonded to a second cover layer, said first and second cover layers each including a plurality of slits disposed to define at least respective hinged flaps, said first layer flap and said second layer flap disposed such that said first layer slits and said second layer slits are not aligned.
Claim 5
Dependent Claim 16 words
The cartridge of claim 3 wherein said first layer slits define a generally y-shaped slit arrangement.
Claim 6
Dependent Claim 61 words
The apparatus of claim 1 wherein said cartridge conveying means comprises rotatable carousel means and wherein said first locking means comprise radial lock means for positioning and locking said cartridge in a radial direction, circumferential lock means for positioning and locking said cartridge in a circumferential direction, and vertical lock means for positioning and locking said cartridge in a vertical direction.
Claim 7
Dependent Claim 149 words
The apparatus of claim 5 wherein said circumferential lock means comprises a radially extending carousel wall having at least one circumferential contact point; circumferential contact point engagement means on said cartridge for engaging said cartridge such that said circumferential contact point engagement means engages such circumferential contact point; said horizontal lock means comprises a surface on said cartridge having at least one horizontal contact point; tab means attached to said carousel for engaging said horizontal contact point; and means for biasing said cartridge such that said horizontal contact point engages said tab means; said radial lock means comprises a circumferentially extending surface on said carousel, said circumferentially extending surface having at least one tangential contact point; tangential contact point engagement means on said cartridge for engaging said tangential contact point., and means for biasing said cartridge such that said tangential contact point engages said tangential contact point engagement means.
Claim 8
Dependent Claim 17 words
The apparatus of claim 1 wherein said second locking means is self-locking with said first locking means.
