Abstract

Abstract A specific binding composition comprises a specific binding species and one or more water-soluble proteins or carbohydrates, substantially none of which has a pI greater than 5. This composition provides improved sensitivity with lower background in diagnostic tests. Preferably, the species is labeled for detection, for example, with an enzyme. The composition can be included with a dye-providing composition in a diagnostic test kit for use in diagnostic methods.

Claims Analysis

Total Claims: 13
Claim 1
Independent Claim 45 words
A specific binding composition comprising a specific binding species and characterized as having one or more water-soluble proteins or carbohydrates, a major proportion by weight of the total protein and carbohydrate consisting of proteins or carbohydrates having a pI equal to or less than 5.
Claim 2
Dependent Claim 45 words
A specific binding composition comprising a specific binding species and characterized as having one or more water-soluble proteins or carbohydrates, a major proportion by weight of the total protein and carbohydrate consisting of proteins or carbohydrates having a pI equal to or less than 5.
Claim 3
Dependent Claim 27 words
The composition as claimed in claim 1 wherein the water-soluble protein or carbohydrate is succinylated casein, glutarylated casein, carboxymethyl cellulose, succinylated bovine serum albumin or succinylated collagen.
Claim 4
Dependent Claim 32 words
The composition as claimed in claim 1 or claim 2 wherein the water-soluble protein or carbohydrate is present in an amount of from 0.05 to 2 percent, based on total composition weight.
Claim 5
Dependent Claim 20 words
The composition as claimed in any one of claims 1 to 3 wherein the specific binding species is an antibody.
Claim 6
Dependent Claim 20 words
The composition as claimed in any one of the preceding claims wherein the specific binding species is labeled for detection.
Claim 7
Dependent Claim 12 words
The composition as claimed in claim 5 wherein the species is enzyme-labeled.
Claim 8
Dependent Claim 22 words
The composition as claimed in any one of claims 1 to 3 wherein the species is an enzyme-labeled antibody to Streptococcus A.
Claim 9
Dependent Claim 43 words
A diagnostic test kit for the detection of a target ligand comprising: a. a specific binding composition as claimed in claim 6 or claim 7, and b. a dye-forming composition which is capable of providing a dye in the presence of the enzyme.
Claim 10
Independent Claim 76 words
A method for the detection of a target ligand in a biological specimen comprising the steps of: A. contacting a sample of a biological specimen suspected of containing a target ligand with a specific binding composition as claimed in any of claims 1 to 7, to form a specific binding complex between the ligand and species, and B. detecting the presence of the complex as an indication of the presence of the ligand in the specimen.
Claim 11
Dependent Claim 30 words
The method as claimed in claim 9 wherein, prior to, simultaneously with or subsequent to contacting step A, contacting the ligand with a receptor for the ligand which is insolubilized.
Claim 12
Dependent Claim 39 words
The method as claimed in claim 9 wherein, prior to contacting step A, the ligand is contacted with an unlabeled receptor therefor, and the specific binding composition comprises an enzyme-labeled specific binding species specifically reactive with the unlabeled receptor.
Claim 13
Independent Claim 19 words
The method as claimed in any of claims 9 to 11 for the determination of a Streptococcus A antigen.
