Abstract
Abstract Highly concentrated hematopoietic stem cell compositions are provided which are substantially free of differentiated or dedicated hematopoietic cells. The cells are obtained by subtraction of cells having particular markers and selection of cells having particular markers. The resulting composition may be used to provide for individual or groups of hematopoietic lineages, to reconstitute stem cells of the host, and to identify an assay for a wide variety of hematopoietic growth factors.

Claims Analysis
Total Claims: 11
Claim 1
Independent Claim
20 words
A cell composition consisting essentially of as the cellular population viable mammalian hematopoietic stem cells from the same mammalian source.
Claim 2
Dependent Claim
20 words
A cell composition consisting essentially of as the cellular population viable mammalian hematopoietic stem cells from the same mammalian source.
Claim 3
Dependent Claim
13 words
A cell composition according to Claim 1, wherein said cells are mouse cells.
Claim 4
Dependent Claim
183 words
A method for preparing a cell composition according to Claim 1, said method comprising: (1) separating a bone marrow cell population into a T-cell population and a substantially T-cell free population by means of T-cell markers; (2) separating a bone marrow cell population into a high level cell differentiation marker containing population and a low level cell differentiation marker containing population; (3) separating a bone marrow cell population into a population substantially free of markers of major cell populations of hematopoietic dedicated cells and cells comprising at least one marker of a hematopoietic dedicated cell; (4) separating a bone marrow cell population substantially free of markers of hematopoietic dedicated cells into cells that bind to an antibody to stem cell antigen and those that do not bind, wherein said cells which bind to stem cell antigen are a substantially homogeneous population of stem cells; with the proviso that steps (1), (2) and (3) may be performed in any order, with the cells from the previous step used in the next successive steps and the resulting bone marrow cell population used in step (4).
Claim 5
Dependent Claim
24 words
A method according to Claim 3, wherein step (1) is performed with magnetic beads and step (4) is performed by fluorescence activated cell sorting.
Claim 6
Dependent Claim
19 words
A method according to Claim 4, wherein in step (3) the separating of said dedicated cells is performed simultaneously.
Claim 7
Dependent Claim
20 words
A method according to Claim 5, wherein said simultaneous separation is performed using magnetic beads of fluorescence activated cell sorting.
Claim 8
Dependent Claim
35 words
A method for producing dedicated hematopoietic cells, said method comprising: contacting stem cells according to Claim 1 with at least one growth factor which results in said stem cells differentiating into a dedicated hematopoietic cell.
Claim 9
Dependent Claim
18 words
A method according to Claim 8, wherein said growth factor results from a medium resulting from stromal cells.
Claim 10
Dependent Claim
41 words
A method for producing a mixture of dedicated hematopoietic cells, said method comprising: contacting stem cells according to Claim 1, with at least one growth factor which results in said stem cells differentiating into at least two different dedicated hematopoietic cells.
Claim 11
Dependent Claim
50 words
A method for assaying for a growth factor capable of causing the dedication of a stem cell to a particular lineage, said method comprising: contacting stem cells according to Claim 1 with a sample suspected of containing said growth factor; and determining the population of dedicated cells of said lineage.
