Claims Analysis
Total Claims: 10
Claim 1
Independent Claim
111 words
A method for amplifying a nucleic acid comprising a target sequence comprising the steps of: a) generating a single-stranded template comprising the target sequence; b) hybridising a pair of oligonucleotides to the template such that the oligonucleotides are juxtaposed for ligation on the template and each oligonucleotide is positioned to flank the target sequence; c) ligating the juxtaposed oligonucleotides to produce a double-stranded ligation product; d) denaturing the double stranded ligation product to produce an initial template strand and an oligonucleotide ligation product strand; and e) repeating steps (b) to (d) such that both the initial template strand and the oligonucleotide ligation strand act as templates for a further ligation reaction.
Claim 2
Claim 3
Dependent Claim
23 words
A method according to claim 1 wherein the single stranded template is generated by disassociating the complementary strands of a double-stranded nucleic acid.
Claim 4
Dependent Claim
14 words
A method according to claim 2 wherein both complementary strands are used as templates.
Claim 5
Independent Claim
17 words
A method according to any one of claims 1 to 3 wherein the nucleic acid is DNA.
Claim 6
Independent Claim
14 words
A method according to claims 1 to 3 wherein the nucleic acid is RNA.
Claim 7
Independent Claim
15 words
A method according to any preceding claim wherein the target sequence is a single nucleotide.
Claim 8
Independent Claim
42 words
A method according to any preceding claim further comprising producing an amplification product by repetition of step (e) and subsequently determining the presence or absence of a nucleic acid sequence complementary to or identical to the target sequence in the amplification product.
Claim 9
Independent Claim
27 words
A method for the diagnosis of a genetic disease comprising analysing the genotype of a patient by the method of any one of claims 1 to 7.
Claim 10
Dependent Claim
13 words
A method according to claim 8 wherein the disease is sickle cell anaemia.
