A highly specific method for detecting nucleic acids in solution
Abstract
For the detection of nucleic acids of a defined sequence by hybridization with two single-stranded nucleic acid probes that are present in the same solution phase and complementary to different areas of the nucleic acid to be detected, one nucleic acid probe serving as a detector probe and containing at least one hapten bound as a label via a chemical compound, and the other nucleic acid probe serves as a capture probe and is bound to a solid matrix, a steroid is used as a hapten, which is covalently bound to at least one position of the detector probe, which is not involved in hydrogen bonding with the nucleic acid to be detected, via a bridge of at least four atoms in length, incubate the nucleic acid to be detected in solution with the detector probe and the capture probe in any order, the binding of the capture probe to the matrix before, during or after the incubation with the nucleic acid to be detected causes, separates the remaining solution from the matrix-bound complex of capture probe, nucleic acid to be detected and detector probe and detects the complex via an anti-hapten antibody labeled in turn.
