Claims Analysis

Total Claims: 10
Claim 1
Independent Claim 64 words
A method of preparing hyaluronic acid comprising the steps of (a) culturing an hyaluronic acid- producing organism which is either hyaluronidase-negative or hyaluronidase-inhibited under conditions sufficient to assure an enhanced amount' of hyaluronic acid in the organism, the culturing being in a medium substantially free of protein; (b) extracting the hyaluronic acid from the culture; and (c) removing reactive proteins via protein binding filtration.
Claim 2
Independent Claim 64 words
A method of preparing hyaluronic acid comprising the steps of (a) culturing an hyaluronic acid- producing organism which is either hyaluronidase-negative or hyaluronidase-inhibited under conditions sufficient to assure an enhanced amount' of hyaluronic acid in the organism, the culturing being in a medium substantially free of protein; (b) extracting the hyaluronic acid from the culture; and (c) removing reactive proteins via protein binding filtration.
Claim 3
Dependent Claim 22 words
The method of claim 1 wherein the organism is selected from the group consisting of group A and group C hemolytic streptococci.
Claim 4
Dependent Claim 11 words
The method of claim 3 wherein the organism is Streptococcus equi.
Claim 5
Dependent Claim 58 words
The method of claim 1 wherein the extraction of step (b) is followed by a purification process comprising the steps of repeated alcohol precipitation and H 2 0 solubilization followed by filtration through nitrocellulose until the protein content is less than about 1.25 mg/ml and the nucleic acid concentration is less than about 45 µg/ml of the preparation.
Claim 6
Dependent Claim 33 words
A sterile preparation comprising hyaluronic acid or a salt thereof, the preparation being substantially free of proteins and nucleic acids and having a single, closely controlled molecular weight range as determined by HPLC.
Claim 7
Dependent Claim 33 words
A sterile preparation comprising hyaluronic acid or a salt thereof, the preparation being substantially free of proteins and nucleic acids and having a single, closely controlled molecular weight range as determined by HPLC.
Claim 8
Dependent Claim 42 words
The preparation of claim 5 wherein the protein content is less than about 1.25 mg/ml, the nucleic acid content is less than about 45 µg/ml and the preparation, when subjected to HPLC, is characterized by an essentially single, substantially symmetrical distribution peak.
Claim 9
Dependent Claim 26 words
The preparation of claim 6 wherein the protein content is less than about 0.10 mg/ml and the nucleic acid content is less than about 5 µg/ml.
Claim 10
Dependent Claim 34 words
The preparation of claim 7 wherein the preparation when used as an equine joint fluid replacement has a Clinical Index of less than 6.0 as determined by the Horse Joint Reactivity Test described herein.
