Abstract

Abstract A method for fusing plant protoplasts comprising the steps of; (a) treating the plant protoplasts with a fusing agent, and then (b) incubating the resultant plant protoplasts in an incubation medium having an osmotic pressure lower that that required for maintaining the plant protoplasts in a stable form. This effectively promotes the fusion of the plant protoplasts at a higher fusion rate in a wider range of plants.

Claims Analysis

Total Claims: 11
Claim 1
Independent Claim 40 words
A method of fusing plant protoplasts comprising treating the plant protoplasts with fusing agent and incubating the resultant plant protoplasts in an incubation medium having an osmotic pressure lower than that required for maintaining the plant protoplasts in stable form.
Claim 2
Independent Claim 40 words
A method of fusing plant protoplasts comprising treating the plant protoplasts with fusing agent and incubating the resultant plant protoplasts in an incubation medium having an osmotic pressure lower than that required for maintaining the plant protoplasts in stable form.
Claim 3
Dependent Claim 23 words
A method according to claim 1 wherein the fusion is carried out between plant protoplasts of the same genus or of different genera.
Claim 4
Dependent Claim 32 words
A method according to claim 2 wherein protoplasts of at least one plant selected from the group consisting of genera Lithospermus, Nicotinia; Coptis, Dianthus, Duboisia Rosa, Oryza, Triticum, and Zea are fused.
Claim 5
Independent Claim 29 words
A method according to any preceding claim wherein the fusing agent comprises at least one of polyethylene glycol, polypropylene glycol, polyvinyl alcohol, dextrin, pectin, calcium chloride, and sodium nitrate.
Claim 6
Independent Claim 24 words
A method according to any preceding claim wherein .the osmotic pressure of the incubation medium is not more than 1 0 kg/cm 2 .
Claim 7
Independent Claim 17 words
A method according to any preceding claim wherein the incubation period is not more than 48 hours.
Claim 8
Independent Claim 31 words
A method according to any preceding claim wherein the incubation medium contains at least one material selected from sucrose, glucose, mannitol, sorbose, calcium chloride, magnesium chloride, nutrients for plant cell culture.
Claim 9
Independent Claim 79 words
A method according to any preceding claim wherein the incubation medium contains at least one fusion promoting agent selected from water-soluble proteins (e.g. albumin, globulin, papain, and bromelin) and hydrophilic colloidal synthetic polymers (e.g. those prepared by coating at least one of polyvinyl pyrrolidone and polyacrylamide on fine particles of at least one insoluble material selected from glass, silica, alumina and active carbons and those which are prepared from sucrose and epichlorohydrin and have a plurality of branched structures).
Claim 10
Independent Claim 40 words
A method according to claim.9 wherein the said protein and/or polymer fusion promoting agent is present in a total amount of 0.001 to 10 parts by weight, per 100 parts by weight of the incubation medium containing the plant protoplast.
Claim 11
Independent Claim 73 words
A method according tp any preceding claim wherein the incubation medium contains at least one fusion promoting agent selected from water-soluble aprotic polar compounds (e.g. sulforane, dimethyl sulfone, dimethyl sulfoxide, diethyl sulfoxide, N,N-dimethylformamide, N,N-diethylformamide, N,N--dimethylacetamide, N-methylpyrrolidone, N-ethylpyrrolidone, N-isopropylpyrrolidone, N-octylpyrrolidone, N-cyclohexylpyrrodone, N-benzylpyrrolidone, polyvinylpyrrolidone, N-methyl-2-pyridine, N-methyloxazolidone, hexamethylphosphoric triamide, and hexaethylphosphoric triamide), preferably in a total amount of 0.01 to 10 parts by volume, per 100 parts by volume of the incubation medium containing the protoplasts.
