Abstract

Abstract A process for carrying out a specific binding assay (for example an immunoassay) in which (a) a sample under assay, possibly containing a substance being tested for, is reacted with (b) a specific binding partner for the substance being tested for, immobilised on a solid support, and (c) a specific binding partner for the substance being tested for which is conjugated to a detectable marker, thereby to form a complex by reaction between whatever quantities are present of the substance being tested for with reagents (b) and (c), in which the marker is immobilised to the support via the substance being tested for, andis detected or assayed as an index of the quantity present in the sample (a) of any of the substance being tested for; characterised in that reaction ingredients (a), (b) and (c) are all mixed in a single step for reaction in a single reaction liquid, and competitive interference between the binding reactions of the substance being tested for and reagents (b) and (c) is avoided either by use of an antibody of narrow and different, non-interfering specificity, such as a monoclonal antibody, to avoid the interference, or by use of a slow-release form of reagent (c).

Claim 3
Dependent Claim 26 words
A process according to claim 1, characterised in that a monoclonal antibody immobilised on a solid surface to form an immunosorbent is comprised in reagent (b).
Claim 4
Dependent Claim 22 words
A process according to claim 1 or 2, characterised in that a monoclonal antibody in marker-conjugated form is .comprised in reagent (c).
Claim 5
Dependent Claim 35 words
A process according to claim 1,2 or 2, characterised in that reagent (c) is used in the reaction in slow-release form such as a glaze coated on a surface in contact with the reaction liquid.
Claim 6
Dependent Claim 51 words
A process according to any of claims 1-4, characterised in that the specific binding reagent (b) is immobilised on the surface of a displacer body (2) which occupies a majority of the volume of a well or cup (1) containing the aqueous liquid in which the specific binding reaction takes place.
Claim 7
Dependent Claim 52 words
A process according to claim 5, in which the displacer body (2) has a shape substantially complementary to and slightly smaller than that of the well or cup (1), preferably reducing the volume available in the well or cup (1) for occupation by the aqueous reaction liquid by a factor of a-10.
Claim 8
Dependent Claim 61 words
A process according to any of claims 1-6, characterised in that the immobilised specific binding reagent (b) is either an immobilised antibody or an immobilised substance to which an immunoglobulin under assay can specifically bind, and marker conjugated-reagent (c) comprises either another antibody specific to the binding partner of antibody reagent (b), or an antiglobulin specific for an immunoglobulin under assay.
Claim 9
Dependent Claim 109 words
A process for carrying out a specific binding assay in which (a) a sample under assay, possibly containing a substance being tested for, is reacted with (b) an antibody of corresponding specificity immobilised on a solid support and (c) an antibody (also with binding specificity for the substance being tested for) conjugated to a detectable marker, thereby to form a complex between whatever quantities are present of the substance being tested for with reagents (b) and (c), and in which after the binding reaction the amount of detectable marker bound is directly or indirectly assayed, characterised in that the antibodies comprised in reagents (b) and (c) are monoclonal antibodies.
Claim 10
Claim 11
