Abstract

Abstract A system and method is disclosed for the rapid detection and (or) purification of analytes in a sample. The system makes use of a fluid receptacle containing a capture reagent and a manifold having two ports. A sample containing an analyte disposed within the fluid receptacle is subjected to a capture reagent and processed by inserting the open end of the receptacle in one port. Vacuum, gas, reagents, and wash fluids are then applied to the second port to effect rapid interaction between the capture reagent and analyte, efficient washing of the capture reagent and detection and/or collection of the analyte. The system and method provide a rapid safe, uncomplicated means of automating processes based on affinity complexation principles. Encompassed within this category of processes are immunoassays, nucleic acid hybridization tests and affinity separations.

Claims Analysis

Total Claims: 7
Claim 1
Independent Claim 109 words
A method for processing a sample having an analyte by the steps of: using a closed receptacle, the receptacle having a single access port positioned below the remainder of the receptacle, together with an immobilized capture reagent for the analyte, and a porous retainer able to pass easily fluids, cells and the like in the receptacle for securing the capture reagent, subjecting the access port and hence the receptacle to an initial vacuum of at least 25 inches of mercury, and thereafter selectively subjecting the access port to reagents, vacuum, air, and/or wash fluids while maintaining a vacuum within the receptacle thereby to capture, wash and develop the analyte.
Claim 2
Claim 3
Dependent Claim 49 words
The method of claim 1 wherein the capture reagent is immobilized on the porous retainer and the receptacle access port is subjected to coordinated application of vacuum and air while retaining a vacuum within the receptacle to recycle the analyte and reagents back and forth across the capture reagent.
Claim 4
Dependent Claim 48 words
The method of claim 1 wherein the second step includes alternately introducing into and removing from the access port air to intermix the sample with the capture reagent while retaining a vacuum and the sample in the receptacle, thereby to capture the analyte on the immobilized capture reagent.
Claim 5
Dependent Claim 92 words
The method of claim 1 wherein the step of selectively subjecting the receptacle includes: (1) subjecting the access port to a vacuum sufficient to remove the sample, (2) while maintaining the access port below atmospheric pressure, washing the immobilized capture reagent by subjecting the access port to alternate bursts of air and wash fluid, (3) subjecting the access port to vacuum to remove the wash fluids, (4) introducing a signal generating reagent to the evacuated receptacle, and thereafter repeating step (2) and allowing the pressure in the receptacle to return to atmospheric.
Claim 6
Dependent Claim 23 words
The method of claim 1 wherein the capture reagent is immobilized on beads in the receptacle which cannot pass through the porous retainer.
Claim 7
Dependent Claim 125 words
The method of claim 1 the second step includes alternately introducing into and removing from the access port air to intermix the sample with the capture reagent while retaining a vacuum and the sample in the receptacle, thereby to capture the analyte on the immobilized capture reagent (1) subjecting the access port to a vacuum sufficient to remove the sample, (2) while maintaining the access port below atmospheric pressure, washing the immobilized capture reagent by subjecting the access port to alternate bursts of air and wash fluid, (3) subjecting the access port to vacuum to remove the wash fluids, (4) introducing a signal generating reagent to the evacuated receptacle, and thereafter repeating step (2) and allowing the pressure in the receptacle to return to atmospheric.
