Claims Analysis

Total Claims: 11
Claim 1
Independent Claim 100 words
A method for determining unsaturated thyroxine binding protein sites in a sample containing thyroxine binding proteins, comprising: (a) treating the sample with a composition comprising a nonfluorescent surfactant in a buffered medium having a pH greater than 7.0; (b) contacting the treated sample with a fluorescent labeled tracer wherein said fluorescent labeled tracer has a preferential binding affinity for thyroxine binding globulin, thereby forming a complex between the thyroxine binding proteins and fluorescent labeled tracer; and (c) determining the amount of complex formed utilizing fluorescence polarization techniques, as a measure of the unsaturated thyroxine binding protein sites in the sample.
Claim 2
Claim 3
Dependent Claim 14 words
A method according to claim 1 wherein the nonfluorescent surfactant is sodium dodecyl sulfate.
Claim 4
Dependent Claim 19 words
A method according to claim 2 wherein the pH of the buffered media is in the range of 8.0-9.0.
Claim 5
Dependent Claim 12 words
A method according to claim 3 wherein the buffer is sodium bicarbonate.
Claim 6
Dependent Claim 16 words
A method according to claim 2 wherein the buffered media comprises triethylenediamine in a phosphate buffer.
Claim 7
Dependent Claim 15 words
A method according to claim 1 wherein the fluorescent labeled tracer comprises a thyroxine-fluorescein conjugate.
Claim 8
Dependent Claim 27 words
A method according to claim 6 wherein the thyroxine-fluorescein conjugate is a compound of the formula: ##STR11## wherein m is an integer of from 2 to 8.
Claim 9
Dependent Claim 16 words
A method according to claim 7 wherein m is an integer of from 2 to 4.
Claim 10
Dependent Claim 10 words
A method according to claim 8 wherein m is 3.
Claim 11
Independent Claim 16 words
A method as in any of claims 1-9 wherein steps (a) and (b) are conducted simultaneously.
