Abstract

Abstract Ampholytes are rapidly separated with high resolution in preparative scale by isoelectric focusing at high power inputs. An improved technique for isoelectric focusing involves sequential treatment of an ampholyte mixture in successively smaller focusing units without substantial dilution. Improved isoelectric focusing devices operating at high power inputs, with highly efficient heat dissipation, are described.

Claims Analysis

Total Claims: 4
Claim 1
Independent Claim 81 words
AN IMPROVED METHOD OF PREPARATIVE ISOELECTRIC FOCUSING OF THE TYPE UTILIZING PASSAGE OF A DIRECT CURRENT THROUGH AN AMPHOLYTE SOLUTION, SAID AMPHOLYTE SOLUTION BEING COMPRISED OF A SAMPLE AMPHOLYTE MIXTURE, A CARRIER AMPHOLYTE MIXTURE AND WATER, SAID CARRIER AMPHOLYTE MIXTURE PROVIDING AN ARTIFICIAL PH GRADIENT, IN A MULIT-CHAMBER ISOELECTRIC FOCUSING DEVICE, WHEREIN THE IMPROVEMENT COMPRISES SEQUENTIALLY SUBJECTING SAID AMPHOLYTE MIXTURE, OR DESIRED PORTIONS THEREOF, TO ISOELECTRIC FOCUSING IN OF MULTI-CHAMBER DEVICES OF SUCCESSIVELY SMALLER VOLUME, WITHOUT SUBSTANTIAL DILUTION OF SAID AMPHOLYTE.
Claim 2
Dependent Claim 81 words
Claim 3
Dependent Claim 98 words
The method of claim 1 wherein said sequential treatment utilizes a series of devices of successively smaller volume, such that each device has the same number of chambers, and the ratio of volume of each device to the next smaller device is approximately equal to the number of chambers in each device, in a manner such that the contents of one chamber of a device after isoelectric focusing are transferred to fill the next smaller device without substantial dilution of said ampholyte solution, and isoelectric focusing is repeated until the desired degree of sample ampholyte separation is achieved.
Claim 4
Dependent Claim 16 words
The method of claim 2 wherein the number of chambers in each device is about 10.
