Cancer antigen
Abstract
Abstract NEW MATERIAL:An cancer antigen, produced by a hybridoma of a B lymphatic cell of an animal immunized with a human cancerous cell and tumorous cell, unreactive with normal cells, recognized by an antihuman cancer monoclonal antibody specifically reactive with cancerous cells of plural organs and capable of exhibiting principal bands of about 45,000, 68,000-92,000 and/or 200,000mol.wt. by sodium dodecyl sulfate (SDS)-polyacrylamide gel electrophoresis (PAGE) and showing affinity for concanavalin A lectin. USE:A diagnostic agent and a raw material for production thereof. PREPARATION:For example, an established cancerous cell TMK-1 is stirred in a buffer solution containing a surfactant in a vortex mixer, etc., to provide a homogenate, which is then centrifuged to afford a supernatant. The resultant supernatant is subsequently purified by DEAE ion exchange column and concanavalin A agarose column, etc., to afford a cancerous antigen.
