Method for measuring enzyme activity using reflection optical system
Abstract
Abstract PURPOSE:To measure an enzyme activity value with high accuracy in a method for measuring enzyme activity using a dry type analyzing element by determining the relation between a reflection optical density value and dye concn. value and making the correlation equation between the fluctuation of the dye concn. with time and the enzyme activity value. CONSTITUTION:The reflection optical density (ODR) of the dry type analyzing element is measured with standard solns. having different concns. [P] of dyes or chemical species which can give the dyes, then the correlation equation [P]=f(ODR) is determined. The reflection optical density is then measured at spaced time intervals at the prescribed temp. in an incubator and the concn. fluctuation DELTA[P] of the dye per unit time is determined. The reflection optical density is measured at spaced time intervals by using the standard soln. of which the enzyme activity is measured by a reference method. The correlation equation [E]=f(DELTA[P]) between the enzyme activity value [E] and the concn. fluctuation DELTA[P] of the dye is made. The reflection optical density of the sample of the enzyme in a vital body is measured by the dry type analyzing element and is substituted into the above-mentioned correlation equation; therefore, the enzyme activity is measured with the high accuracy.
