Preventing agent for dental caries
Abstract
Abstract PURPOSE:A preventing agent, containing indigenous bacteria in oral cavity obtained by introducing a gene DNA of alpha-1,3-glucanase or/and alpha-1,6 glucanase into cells for gene expression and capable of preventing dental caries surely and for a long period. CONSTITUTION:A bacterium capable of producing alpha-1,3 glucan 3- glucanohydrolase (alpha-1,3 glucanase) or/and alpha-1,6 glucan 6-glucanohydrolase (alpha-1,6 glucanase), e.g. Bacillus circulans BC-8 species, is cultivated to extract the gene DNA of glucanase in the above-mentioned bacterium. The extracted DNA is then integrated into plasmid DNA of Escherichia coli to give a recombinant plasmid DNA, which is then introduced into Escherichia coli to carry out gene expression. After confirming the presence of the glucanase, gene DNA of the glucanase in the above-mentioned plasmid DNA is integrated into a plasmid DNA extracted from indigenous bacteria in the oral cavity, e.g. Streptococcus sanguis, to make the above-mentioned bacteria carry out gene expression. The resultant bacteria are then applied to teeth in the oral cavity.
