Abstract

Abstract PURPOSE:To determine amylase activity, by adding amylase to a solution of a bonded alpha-glucoside having a polymerization degree of >=5 as a substrate, and measuring the change in the optical rotation of the substrate solution caused by the hydrolysis of the bonded alpha-glucoside. CONSTITUTION:A solution containing a bonded alpha-glucoside having a polymerization degree of >=5 as substrate, preferably maltopentaose, is added with an amylase as a specimen together with glucosidase and mutarotase as assistant enzymes. The change in the optical rotation of the solution is measured by using e.g. Na-D line of 589nm wavelength.
