Rapid detecting method of microorganism
Abstract
Abstract PURPOSE: To measure or detect a microorganism decomposing lactose in a specimen rapidly and at a high sensitivity, by using 4-methyl-umbelliferyl-β-galactoside (4-MUD). CONSTITUTION: A given amount of a specimen is reacted with a solution of 4-MUD (4-methyl-umbelliferyl-β-D-galactoside) or added to a nutrient culture medium containing a β-galactosidase inducer and/or deoxycholic acid or bile acid and cultivated for 2hr or longer and then reacted with a solution of 4-MUD to destroy or dissolve the microbial cell in the culture fluid to be tested, preferably prior to or during the reaction. β-Galactosidase is thus eluted from the microbial cell to measure the 4-methylumbelliferone produced by the reaction of the β-galactosidase with the 4-MUD.
