Production of indolecarboxylic acid derivative
Abstract
Abstract PURPOSE: To advantageously obtain the subject compd. by an one-shot process by reacting an aniline deriv. with sodium nitrite in an acidic medium under specific temp. conditions to reduce the same by metal zinc and reacting the reduced product with lower alkyl private. CONSTITUTION: A compd. represented by formula I (wherein R 2 is lower alkyl or carbocyclic aryl; and R 3 and R 4 are each H, lower alkyl or lower alkoxy) is reacted with sodium nitrite in an acidic medium such as a liquid org. acid at 0-30°C to obtain a compd. represented by formula II. Subsequently, metal zinc is added to the reaction soln. in an amt. twice the compd. represented by the formula II so that reaction temp. is held to 0-30°C reduce the compd. in the presence of a sufficient amt. of an acid to obtain a compd. represented by formula III. Further, the reaction soln. is reacted with a compd. represented by formula IV (R 1 ' is lower alkyl) at a temp. from room temp. to 120°C and the formed compd. represented by the formula V is succeeedingly hydrolyzed if desired to directly obtain the objective compd. represented by formula V1 (R 1 is H or R 1 ')from the raw material without isolating the intermediate.
