Immunological treatment of tumor using anti-idiotype antibody
Abstract
Abstract PURPOSE: To enable utilization of an anti-idiotypic antibody for immunotherapy and immunoprophylaxis of tumors by proliferating a hybridoma obtained by fusing a specific cell to a cell of a myeloma. CONSTITUTION: (A) An antibody-producing cell is obtained by inducing thereof from the spleen of an animal immunized with an antibody or a T cell having an idiotype recognizing a human melanoma-associated ganglioside GD3 antigen or GD3 antigen. The resultant cell A is then fused to (B) a cell of a myeloma to afford (C) a hybridoma such as ATCC NaHB9484. The component C is subsequently injected into a pristane-primed mouse to produce an ascitic tumor and grow the tumor. Thereby, (D) a monoclonal antibody is secreted in the ascites fluid. The resultant antibody D is purified by an affinity chromatograph on a protein A Sepharose (R) or by an (NH 4 ) 2 SO 4 precipitation method to produce (E) an anti-idiotypic antibody. The antibody E or its fragment or a modification thereof is formulated so as to provide a therapeutically effective amount of a preparation. Thereby, an immunotherapeutic agent for tumors is produced. COPYRIGHT: (C)1990,JPO
