Abstract
Abstract An immunoassay method and apparatus in which scattered light is measured, said light source being aligned to direct illumination toward an interface between a sample container and an aqueous solution at an angle less than the critical angle. A detector to measure the scattered light is located at a place outside the envelope of the critical angle.

Total Claims: 12
Claim 1
Independent Claim
123 words
An apparatus for conducting an immunoassay to detect a ligand labelled with light scattering particles or a ligand binding partner labelled with light scattering particles in an aqueous solution comprising: (a) a receiving means for receiving and holding an aqueous solution comprising at least one portion consisting of an optically transmissive material, said material having a refractive index greater than that of said aqueous solution; (b) illumination means for providing light to an interface formed between said optically transmissive material portion of the receiving means and an aqueous solution in said receiving means; and (c) photodetector means for receiving light scattered by said light scattering particles; wherein the light is directed to strike the interface at an angle less than the critical angle.
Claim 2
Dependent Claim
21 words
The apparatus as recited in claim 1, wherein said photodetector means is located in a position to detect forward scattered light.
Claim 3
Dependent Claim
17 words
The apparatus as recited in claim 1, wherein said optically transmissive receiving means is a sample container.
Claim 4
Dependent Claim
16 words
The apparatus as recited in claim 1, wherein said optically transmissive receiving means is a cuvette.
Claim 5
Dependent Claim
14 words
The apparatus as recited in claim 1, wherein said illumination means is a laser.
Claim 6
Dependent Claim
36 words
The apparatus as recited in claim 1, further comprising a light directing means wherein the illumination is directed to the interface between said optically transmissive receiving means and said aqueous solution by a light directing means.
Claim 7
Dependent Claim
15 words
The apparatus as recited in claim 6, wherein said light directing means is a prism.
Claim 8
Dependent Claim
39 words
The apparatus as recited in claim 1, wherein said optically transmissive receiving means is a slide with said aqueous solution on one surface of said slide, and wherein said illumination is directed through said slide from the other side.
Claim 9
Independent Claim
238 words
A method for determining the presence of a ligand in an aqueous solution comprising the steps of: (a) providing a first ligand binding partner and a second ligand binding partner that have binding affinity for a ligand, wherein at least one of said first ligand binding partner and said second ligand binding partner have binding affinity specific for said ligand, said first ligand binding partner being labeled with a particle having light scattering characteristics in said aqueous solution, and said second ligand binding partner being bound to at least one surface of an aqueous solution receiving means; (b) combining said first ligand binding partner, said aqueous solution and the surface of the solution receiving means having the second binding partner bound thereto said surface being composed of an optically transmissive material with a refractive index greater than the refractive index of said aqueous solution, whereby the second binding partner bound to the surface of the receiving means will bind the ligand and the first binding partner will bind to the ligand to form a light scattering layer; (c) illuminating the receiving means and the aqueous solution at the interface thereof through said optically transmissive material at an angle less than the critical angle; and (d) measuring the amount of light scattered from said interface by the light scattering layer, wherein the amount of scattered light is a function of the amount of ligand present in said aqueous solution.
Claim 10
Dependent Claim
26 words
The method as recited in claim 9, wherein said amount of scattered light measured is light scattered from said interface at greater than the critical angle.
Claim 11
Dependent Claim
21 words
The method as recited in claim 9, wherein said labelled first ligand binding partner is a gold sol labelled monoclonal antibody.
Claim 12
Dependent Claim
21 words
The apparatus as recited in claim 1, wherein said photodetector means is located in a position to detect back scattered light.
