Abstract
Abstract A DNA library comprising the variable region of immunoglobulin light chains and a DNA library comprising the variable region of immunoglobulin heavy chains are obtained. A prokaryotic cell is transformed with plasmid expression vectors containing one of said DNA libraries and the transformed cell is then infected with bacteriophage expression vectors containing the other of said DNA libraries. Infected and transformed cells express proteins. Proteins, including novel proteins, which bind to particular moieties can be selected by screening Fab proteins expressed by infected and transformed cells.

Claims Analysis
Total Claims: 8
Claim 1
Independent Claim
106 words
A method for selecting DNA sequences which encode proteins containing at least two different subunits and which are capable of binding to a particular moiety comprising the steps of: a) transforming a prokaryotic cell with a population of plasmid expression vectors containing a first DNA library; b) infecting said transformed cell with a population of bacteriophage expression vectors containing a second DNA library; wherein one of said DNA libraries comprises the variable region of immunoglobulin light chains and the other of said DNA libraries comprises the variable region of immunoglobulin heavy chain; and c) identifying infected, transformed cell which express proteins that bind to said moiety.
Claim 2
Dependent Claim
57 words
The method of claim 1 wherein the infected, transformed cell are incubated to express proteins comprising at least one subunit encoded by DNA contained in one of the two DNA libraries and at least one subunit encoded by DNA contained in the other of the two DNA libraries, where the subunits have been associated in random assortment.
Claim 3
Dependent Claim
27 words
The method of claim 1 wherein the plasmid contains the DNA library for immunoglobulin light chain and the bacteriophage contains the DNA library for immunoglobulin heavy chain.
Claim 4
Dependent Claim
31 words
The method of claim 1 wherein said DNA library comprising the variable region of the immunoglobulin light chain is obtained by amplifying the rearranged variable region in DNA isolated from lymphocytes.
Claim 5
Dependent Claim
31 words
The method of claim 1 wherein said DNA library comprising the variable region of the immunoglobulin heavy chain is obtained by amplifying the rearranged variable region in DNA isolated from lymphocytes.
Claim 6
Dependent Claim
47 words
The method of claim 1 wherein both said expression vectors also contain DNA sequences encoding an immunoglobulin constant region, where the light chain library is in translational frame with light chain constant sequences and the heavy chain library is in translational frame with heavy chain constant sequences.
Claim 7
Dependent Claim
52 words
The method of claim 1 wherein step (c) further comprises the steps of: a) growing the infected, transformed cell under conditions which induce expression of the proteins encoded by said expression vectors; b) determining which cell express proteins which bind to said moiety; c) recovering DNA from cells identified in step b.
Claim 8
Dependent Claim
9 words
The cell identified in the method of claim 1.
