Abstract

Abstract Antibody complexes comprising hapten-modified diagnostic or therapeutic agents and anti-hapten antibodies are disclosed. Administration of hapten-modified agents, complexed with suitable anti-hapten antibodies, extends the serum half-life and permits an increased concentration of such diagnostic or therapeutic agents at in vivo target sites.

Claims Analysis

Total Claims: 11
Claim 1
Independent Claim 43 words
A process for preparing a pharmaceutical composition which comprises complexing a hapten-modified diagnostic or therapeutic agent with an anti-hapten antibody, said anti-hapten antibody being selected to bind said hapten-modified agent in a manner which does not substantially impair the activity of said agent.
Claim 2
Dependent Claim 43 words
A process for preparing a pharmaceutical composition which comprises complexing a hapten-modified diagnostic or therapeutic agent with an anti-hapten antibody, said anti-hapten antibody being selected to bind said hapten-modified agent in a manner which does not substantially impair the activity of said agent.
Claim 3
Dependent Claim 23 words
A process for preparing a composition as claimed in claim 1 in which said anti-hapten antibody is a monoclonal antibody or fragment thereof.
Claim 4
Dependent Claim 41 words
A process for preparing a composition as claimed in claim 2 wherein said anti-hapten antibody comprises an antibody fragment selected from the group consisting of Fab, Fab', F(ab')₂ fragments or an antibody fragment retaining the essential binding function of said antibody.
Claim 5
Dependent Claim 44 words
A process for preparing a composition as claimed in claim 2 in which said anti-hapten antibody is a chimeric antibody or hybrid monoclonal antibody having a dual specificity, one of which is directed against said hapten-modified agent and the other against a target antigen.
Claim 6
Dependent Claim 27 words
A process for preparing a composition as claimed in claim 4 in which said target antigen is a tumor-associated antigen and said hapten-modified agent has anti-tumor activity.
Claim 7
Dependent Claim 35 words
A process for preparing a composition as claimed in claim 5 in which said target antigen is selected from the group consisting of CEA, PAP, PSA, HCG, ferritin, bombesin, and melanoma-associated antigens p97 and gp240.
Claim 8
Dependent Claim 33 words
A process for preparing a composition as claimed in claim 1 wherein said hapten-modified agent is selected from the group consisting of drugs, toxins, proteins, and biological response modifiers or radio-labeled derivatives thereof.
Claim 9
Dependent Claim 29 words
A process for preparing a composition as claimed in claim 7 in which said hapten-modified agent is a drug capable of binding to a receptor at said target site.
Claim 10
Dependent Claim 29 words
A process for preparing a composition as claimed in any of claims 1 to 8 in which said hapten-modified agent and said anti-hapten antibody are complexed in vitro .
Claim 11
Dependent Claim 30 words
A process for preparing a composition as claimed in claim 1 in which said hapten-modified agent comprises mitomycin C aminobenzyl EDTA thiourea ("MAT"), a metal complex or a derivative thereof.
