Abstract

Abstract The invention relates to DNA sequences which code for the amino-acid sequence of the thrombin inhibitor hirudin, to hybrid vectors containing such DNA sequences, to host cells transformed with such hybrid vectors, to polypeptides produced by such transformed host cells and having thrombin inhibitory activity, to processes for preparing these DNA sequences, hybrid vectors and transformed host cells and processes for preparing these thrombin inhibitors using the transformed host cells. The hirudin compounds have valuable pharmacological properties.

Claims Analysis

Total Claims: 11
Claim 1
Independent Claim 41 words
An expression vector that contains a DNA sequence coding for desulphatohirudin of the formula which sequence is regulated by an expression control sequence in such a manner that the said desulphatohirudin is expressed in a microorganism transformed with that expression vector.
Claim 2
Dependent Claim 21 words
An expression vector according to claim 1, in which the expression control sequence is that of the E. coli tryptophan operon.
Claim 3
Dependent Claim 20 words
An expression vector according to claim 1, in which the expression control sequence is that of the yeast- PHO5 gene.
Claim 4
Dependent Claim 56 words
A process for the manufacture of an expression vector according to any one of claims 1 to 3, characterised in that a DNA sequence coding for desulphatohirudin of the formula is inserted in such a manner into a vector DNA that contains an expression control sequence that the expression control sequence regulates the said DNA sequence.
Claim 5
Dependent Claim 18 words
A microorganism that is transformed with an expression vector according to any one of claims 1 to 3.
Claim 6
Dependent Claim 25 words
A transformed microorganism according to claim 5, wherein the microorganism is selected from a strain of Escherichia coli , Bacillus subtilis or Saccharomyces cerevisiae .
Claim 7
Dependent Claim 9 words
A transformed Escherichia coli strain according to claim 5.
Claim 8
Dependent Claim 9 words
A transformed Saccharomyces cerevisiae strain according to claim 5.
Claim 9
Dependent Claim 34 words
A process for the manufacture of a transformed microorganism according to any one of claims 5-8, characterised in that a microorganism is transformed with an expression vector according to any one of claims 1-3.
Claim 10
Dependent Claim 68 words
A process for the manufacture of biologically active desulfatohirudin of the formula and salts thereof, characterised in that microorganisms transformed with an expression vector that contains a DNA sequence coding for the said hirudin compound and regulated by an expression control sequence are cultivated in a liquid nutrient medium that contains assimilable carbon and nitrogen sources, and the said desulphatohirudin is freed from the host cells and isolated.
Claim 11
Dependent Claim 30 words
A process according to claim 10, characterised in that a free desulphatohirudin obtained is converted into a salt thereof or a desulphatohirudin salt obtained is converted into the free compound.
