Use of 4-amino-N-(1-azabicyclo[2.2.2]oct-3-yl)-5-chloro-2-methoxybenzamide for the manufacture of a medicament having anti-emetic activity.

Claim 2

The use of a 2-alkoxy-N-(1-azabicyclo[2.2.2]-oct-3-yl)benzamide or thiobenzamide of general formula I: wherein: X represents oxygen or sulphur, R 1 represents loweralkyl, R 2 represents hydrogen, halo, 4,5-benzo, alkoxy, amino, methylamino or dimethylamino, n is 1 or 2, or a pharmaceutically acceptable acid addition salt thereof, in the manufactureof a medicament having anti- emetic activity.

The use of 4-amino-N-(1-azabicyclo[2.2.2]oct-3-yl)-5-chloro-2-methoxybenzamide, N-(1-azabicyclo[2.2.2]oct-3-yl)-5-chloro-2-methoxy-4-methylaminobenzamide, N-(1-azabicyclo[2.2.2]oct-3-yl)-2-methoxybenzamide, N-(1-azabicyclo[2.2.2]oct-3-yl)-2,4-dimethoxybenzamide, N-[1-azabicyclo[2.2.2]oct-3-yl]-2-propoxybenzamide, 4-amino-N-(1-azabicyclo[2.2.2]oct-3-yl)-5-chloro-2-methoxybenzamide, N-(1-azabicyclo[2.2.2]oct-3-yl)-5-chloro-2-methoxy-4-methylaminobenzamide, or N-(1-azabicyclo[2.2.2]oct-3-yl)-3-methoxy-2-naphthalenecarboxamide, or a pharmaceutically acceptable acid addition salt thereof, in the manufacture of a medicament having antiemetic activity.

The use of 4-amino-N-(1-azabicyclo[2.2.2]oct-3-yl)-5-chloro-2-methoxybenzamide, N-(1-azabicyclo[2.2.2]oct-3-yl)-5-chloro-2-methoxy-4-methylaminobenzamide, N-(1-azabicyclo[2.2.2]oct-3-yl)-2-methoxybenzamide, N-(1-azabicyclo[2.2.2]oct-3-yl)-2,4-dimethoxybenzamide, N-[1-azabicyclo[2.2.2]oct-3-yl]-2-propoxybenzamide, 4-amino-N-(1-azabicyclo[2.2.2]oct-3-yl)-5-chloro-2-methoxybenzamide, N-(1-azabicyclo[2.2.2]oct-3-yl)-5-chloro-2-methoxy-4-methylaminobenzamide, or N-(1-azabicyclo[2.2.2]oct-3-yl)-3-methoxy-2-naphthalenecarboxamide, or a pharmaceutically acceptable acid addition salt thereof, in the manufacture of a medicament having antiemetic activity.

The use of a 2-alkoxy-N-(1-azabicyclo[2.2.2]-oct-3-yl)benzamide of general formula Ic: wherein: Am represents amino, methylamino or dimethylamino, or a pharmaceutically acceptable acid addition salt thereof, in the manufacture of a medicament having anti- emetic activity.

The use of a 2-alkoxy-N-(1-azabicyclo[2.2.2]-oct-3-yl)benzamide of general formula Ic: wherein: Am represents amino, methylamino or dimethylamino, or a pharmaceutically acceptable acid addition salt thereof, in the manufacture of a medicament having anti- emetic activity.

A pharmaceutical composition comprising a platinum anti-cancer drug and an emesis inhibiting effective amount of a 2-alkoxy-N-(1-azabicyclo[2.2.2]-oct-3-yl)benzamide or thiobenzamide of general formula (I) wherein: X represents oxygen or sulphur, R 1 represents loweralkyl, R 2 represents hydrogen, halo, 4,5-benzo, alkoxy or Am wherein Am represents amino, methylamino or dimethylamino, n is 1 or 2, or a pharmaceutically acceptable acid addition salt thereof.

A pharmaceutical composition comprising a platinum anti-cancer drug and an emesis inhibiting effective amount of a 2-alkoxy-N-(1-azabicyclo[2.2.2]-oct-3-yl)benzamide or thiobenzamide of general formula (I) wherein: X represents oxygen or sulphur, R 1 represents loweralkyl, R 2 represents hydrogen, halo, 4,5-benzo, alkoxy or Am wherein Am represents amino, methylamino or dimethylamino, n is 1 or 2, or a pharmaceutically acceptable acid addition salt thereof.

A pharmaceutical composition comprising a platinum anti-cancer drug and 4-amino-N-(1-azabicyclo[2.2.2]oct-3-yl)-5-chloro-2-methoxybenzamide, or N-(1-azabicyclo[2.2.2]oct-3-yl)-5-chloro-2-methoxy-4-methylaminobenzamide, N-(1-azabicyclo[2.2.2]oct-3-yl)-2,4-dimethoxybenzamide, N-[1-azabicyclo[2.2.2]oct-3-yl]-2-propoxybenzamide, or N-(1-azabicyclo[2.2.2]oct-3-yl)-3-methoxy-2-naphthalene-carboxamide, or a pharmaceutically acceptable salt thereof.

A pharmaceutical composition comprising a platinum anti-cancer drug and 4-amino-N-(1-azabicyclo[2.2.2]oct-3-yl)-5-chloro-2-methoxybenzamide, or N-(1-azabicyclo[2.2.2]oct-3-yl)-5-chloro-2-methoxy-4-methylaminobenzamide, N-(1-azabicyclo[2.2.2]oct-3-yl)-2,4-dimethoxybenzamide, N-[1-azabicyclo[2.2.2]oct-3-yl]-2-propoxybenzamide, or N-(1-azabicyclo[2.2.2]oct-3-yl)-3-methoxy-2-naphthalene-carboxamide, or a pharmaceutically acceptable salt thereof.

A product containing a platinum anti-cancer drug and an antiemetic effective amount of 2-alkoxy-N-(1-azabicyclo[2.2.2]oct-3-yl)benzamide or thiobenzamide of general formula (I) wherein: X represents oxygen or sulphur, R 1 represents loweralkyl, R 2 represents hydrogen, halo, 4,5-benzo, alkoxy or Am wherein Am represents amino, methylamino or dimethylamino, n is 1 or 2, or a pharmaceutically acceptable acid addition salt thereof, for use as a combined preparation for simultaneous, separate or sequential use in cancer therapy.

A process for preparing a 2-alkoxy-N-(1-azabicyclo[2.2.2]oct-3-yl)benzamide or thiobenzamide of general formula I: wherein: X represents oxygen or sulphur; R 1 represents loweralkyl, R 2 represents hydrogen, halo, 4,5-benzo, alkoxy, amino, methylamino or dimethylamino, n is 1 or 2 or a pharmaceutically acceptable acid addition salt thereof comprising either A. where X represents oxygen, either (i) reacting a compound of formula A(i) wherein R 1 , R 2 and n are as defined in formula I with the proviso that when R 2 represents a normally unprotected amino group in formula I it is protected in formula A(i), with 3-aminoquinuclidine; or (ii) reacting a compound of formula A(ii) wherein R 1 , R 2 and n are as defined in formula I with 1,1'-carbonyldiimidazole and 3-aminoquinuclidine; (iii) and optionally converting a compound of formula I so formed into another compound of formula I; or B. where X represents sulphur, (i) reacting a compound of formula I as formed in A above with a mixture of phosphorus pentasulphide and potassium sulphide; or (ii) reacting a compound of formula B(ii) with 3-aminoquinuclidine and sulphur; (iii) and optionally converting a compound of formula I so formed into another compound of formula I.

A process for preparing a 2-alkoxy-N-(1-azabicyclo[2.2.2]oct-3-yl)benzamide or thiobenzamide of general formula I: wherein: X represents oxygen or sulphur; R 1 represents loweralkyl, R 2 represents hydrogen, halo, 4,5-benzo, alkoxy, amino, methylamino or dimethylamino, n is 1 or 2 or a pharmaceutically acceptable acid addition salt thereof comprising either A. where X represents oxygen, either (i) reacting a compound of formula A(i) wherein R 1 , R 2 and n are as defined in formula I with the proviso that when R 2 represents a normally unprotected amino group in formula I it is protected in formula A(i), with 3-aminoquinuclidine; or (ii) reacting a compound of formula A(ii) wherein R 1 , R 2 and n are as defined in formula I with 1,1'-carbonyldiimidazole and 3-aminoquinuclidine; (iii) and optionally converting a compound of formula I so formed into another compound of formula I; or B. where X represents sulphur, (i) reacting a compound of formula I as formed in A above with a mixture of phosphorus pentasulphide and potassium sulphide; or (ii) reacting a compound of formula B(ii) with 3-aminoquinuclidine and sulphur; (iii) and optionally converting a compound of formula I so formed into another compound of formula I.
