Oligonucleotide polymeric support system.
Abstract
A versatile polymer support system for the synthesis of oligonucleotides comprises a universal initiator allowing the extension of a chain, from the 3 ′ or 5 ′ position, with any of the methods of DNA synthesis or commonly available RNA, using a process using oxidative cleaved initiators to facilitate chain elongation and release. The support system can withstand slightly basic and acidic reaction conditions, while allowing an advantageous and quantitative release, before or after removal of the protective groups from the reactive groups, of the oligonucleotides synthesized from a single polymer support. The elimination of the protective groups before the cleavage of the oligomer from the support makes it possible to use the immobilized oligomer as a support for hybridization by affinity both for the isolation and for the detection of complementary polynucleic acids.
