Abstract
Abstract A method for stably binding antigens and allergens to a polystyrene supports, which consists in reacting a polyfunctional aldehyde with the polystyrene solid phase; in washing said solid phase to remove the excess aldehyde; in incubating the so processed solid phase with the antigens or the allergens, and in stabilizing the aldehyde-antigens or the aldehyde-allergens bond with sodium borohydride of with sodium cyanoborohydride.

Claims Analysis
Total Claims: 15
Claim 1
Independent Claim
84 words
A method for stably binding antigens and allergens to solid supports, particularly to polystyrene supports, said method being characterized in that it consists of the steps of - reacting a polyfunctional aldehyde with the solid phase of the support; - removing the excess aldehyde through washing; - incubating the solid phase so treated with the antigens or the allergens; - stabilizing the bond between the aldehyde and the antigens or the allergens by treatment with compounds which are able to reduce the imino-aldehydic bond.
Claim 2
Independent Claim
84 words
A method for stably binding antigens and allergens to solid supports, particularly to polystyrene supports, said method being characterized in that it consists of the steps of - reacting a polyfunctional aldehyde with the solid phase of the support; - removing the excess aldehyde through washing; - incubating the solid phase so treated with the antigens or the allergens; - stabilizing the bond between the aldehyde and the antigens or the allergens by treatment with compounds which are able to reduce the imino-aldehydic bond.
Claim 3
Dependent Claim
20 words
A method according to claim 1, in which the imino- aldehydic bond reducing compound is sodium borohydride or sodium cyanoborohydride.
Claim 4
Dependent Claim
16 words
A method according to claim 1 or 2, in which the polyfunctional aldehyde is the glutaraldehyde.
Claim 5
Dependent Claim
21 words
A method according to claim 1 or 2, in which the polyfunctional aldehyde has from 4 up to 12 carbon atoms.
Claim 6
Independent Claim
22 words
A method according to any of the precedent claims, wherein the solid support is in the form of spheres or of microplates.
Claim 7
Dependent Claim
14 words
A method according to claim 5 wherein the solid support has a rough surface.
Claim 8
Independent Claim
32 words
A method according to any of the precedent claims, wherein the concentration of the polyfunctional aldehyde ranges from 0.01 M to 1 M, and preferably it is between 0.1 and 0.5 M.
Claim 9
Dependent Claim
28 words
A method according to claim 7, wherein the polyfunctional aldehyde is dissolved in a sodium borate or sodium phosphate buffer solution at a pH between 7.1 and 9.
Claim 10
Independent Claim
35 words
A method according to any of the precedent claims, wherein the excess polyfunctional aldehyde is removed after reaction by washing with the same buffer solution as that employed for the preparation of the aldehyde solution.
Claim 11
Independent Claim
29 words
A method according to any of the precedent claims, wherein an inert protein, or amino acids or organic amines are added to the solution containing the antigens or allergens.
Claim 12
Independent Claim
44 words
A method according to any of the precedent claims, wherein the stabilizing treatment is carried out by a sodium borohydride solution that has a concentration between 0.1 and 1 g/1, a pH value between 7.1 and 8, and for a period of some hours.
Claim 13
Independent Claim
34 words
A method according to any of the precedent claims, wherein said support is successively treated with a buffer solution containing an inert protein such as human albumin, bovine serum albumin or horse serum albumin.
Claim 14
Dependent Claim
43 words
A polystyrene solid support containing the antigens or the allergens bound to it in a stable and mechanical/ heat stresses resisting way, as well as in a specific manner, said support being obtained according to the method of any of the precedent claims.
Claim 15
Dependent Claim
42 words
The use of the solid support according to claim 13 for the detection of the IgE, IgG, IgA or IgM classes antibodies specific for antigens or allergens either to diagnose an allergic or an infectious disease, or to check an immunity state.
