Abstract

Abstract A field immunoassay reaction system and method is provided. The reaction system comprises the following components: (i) a dipstick carrier or probe at least partly coated with monoclonal antibody specific for an antigen contained in a body substance or fluid; (ii) a plurality of containers; (iii) detergent or other suitable substance to prevent non-specific binding of antigen to the monoclonal antibody; (iv) buffer; and (v) in situ signal amplification or detection means. Preferably there is also included as an additional component a second monoclonal antibody labelled with an enzyme and enzyme substrate. The detection means is suitably a colour standard or portable colourimeter.

Claims Analysis

Total Claims: 12
Claim 1
Independent Claim 63 words
1, A field immunoassay reaction system for "in situ" testing including: a dipstick probe or carrier at least partly coated with monoclonal antibody specific for an antigen contained in a body substance or fluid; a plurality of containers; detergent or other suitable substance to prevent non specific binding of antigen to the monoclonal antibody; buffer and "in situ" signal amplification or detection means.
Claim 2
Dependent Claim 63 words
1, A field immunoassay reaction system for "in situ" testing including: a dipstick probe or carrier at least partly coated with monoclonal antibody specific for an antigen contained in a body substance or fluid; a plurality of containers; detergent or other suitable substance to prevent non specific binding of antigen to the monoclonal antibody; buffer and "in situ" signal amplification or detection means.
Claim 3
Dependent Claim 22 words
A reaction system as claimed in claim 1 further including a second monoclonal antibody labelled with an enzyme and an enzyme substrate.
Claim 4
Dependent Claim 24 words
A reaction system as claimed in claim 2 wherein the enzyme is a peroxidase and hydrogen peroxide is also included as a co factor.
Claim 5
Dependent Claim 27 words
A reaction system as claimed in any preceding claim wherein at least some of the plurality of containers have identifying means associated therewith to identify individual containers.
Claim 6
Dependent Claim 25 words
A reaction system as claimed in claim 4 wherein there is provided one or more containers for carrying out washing steps in the field assay.
Claim 7
Dependent Claim 19 words
A reaction system as claimed in any preceding - claim wherein there is provided one or more loop dilutors.
Claim 8
Dependent Claim 20 words
A reaction system as claimed in any preceding claim wherein there is also provided a colour standard or portable colourimeter.
Claim 9
Dependent Claim 35 words
E. A reaction system as claimed in any preceding claim wherein the second monoclonal antibody is freezed dried and one of the containers contains diluent for said second monoclonal antibody to reconstitute same when required.
Claim 10
Dependent Claim 100 words
A method of field immunoassay using the reaction system of claim 1 including the steps of: (i) transferring sample to be assayed into a container; (ii) inserting the dipstick probe into the container having the sample through an open top thereof; (iii) removing the probe from the sample container and subsequently washing the probe in one or more containers containing diluent wherein the probe is inserted into said one or more containers through an open top thereof and is subsequently removed; (iv) detecting the presence of an antigen in the sample by subjecting the probe to a signal amplification step.
Claim 11
Dependent Claim 59 words
A method as claimed in claim 9 including the further steps of - (v) inserting the dipstick probe after step (iii) into a container containing enzyme labelled second monoclonal antibody; (vi) washing the dipstick probe in one or more containers containing diluent; and (vii) inserting the dipstick probe into a container containing enzyme substrate before carrying out step (iv).
Claim 12
Dependent Claim 50 words
A dipstick probe or carrier at least partly coated with monoclonal antibody (Mab) specific for an antigen contained in a body substance or fluid, suitable for use in the field immunoassay reaction system of any one of claims 1 to 8 and/or in the method of claim 9 or 10.
